CHEMBIO 3L03 Lecture 8: 2016_Lab #8 ANS Displacment Assay

76 views12 pages

Document Summary

An ans displacement assay will be used to evaluate the ability of your synthesized anthraquinone derivative (lab #3) to interact with purified actr (lab #7). You will share your data with the rest of the class, and determine structure-activity relationships (sar) for the synthesized compounds. Ligand binding underpins most biological processes (e. g. , signal transduction, enzymatic reactions, antibody-antigen interactions). The interaction of a ligand with a protein may occur through a combination of hydrophobic, electrostatic and hydrogen bond interactions. Ligands are extremely diverse and include such molecules as organic compounds (e. g. , metabolic intermediates, amino acids), metal ions or protons, non-metal inorganic ions (e. g. , phosphate), carbohydrates, lipids, and nucleic acids. There are many methods to quantitatively analyze ligand-binding processes (e. g. , fluorescence, radiolabelled ligands, equilibrium dialysis, ultrafiltration, gel filtration). The most useful method distinguishes between the protein-ligand complex and either the free ligand or the free protein without having to separate the components.
