Alzheimer’s Disease

Experiment:
The researchers hypothesized that intravenously or intracerebrally transplanted hASCs
(human adipose derived stem cells) would rescue memory deficit and neuropathology by
up-regulating IL-10 and VEFG therefore, preventing and treating Alzheimer’s disease
(Kim, et al., 2012). The experiment involved the injection of hASCs using a mouse
model in the following methods: intra-venous, which is targeted to prevent the onset of
the disease, and intra-cerebral, which is intended to examine the therapeutic potential in
early stages of the disease (Kim, et al., 2012). The hASCs were intravenously
transplanted into mice biweekly a total of 13 times from 3 months of age or
intracerebrally injected into the gyrus of the hippocampus at 11-months old (Kim, et al.,
2012).
Methods:
Only male animals were used for the purpose of this experiment and divided into four
groups following a polymerase chain reaction. The researchers first isolated and cultured
hASCs from the lower abdomen of patients via liposuction, which were then tested for
cell viability and fungal, bacterial and mycoplasma (bacteria that lack a cell wall around
their membrane) contamination. The hASCs were then stained with fluorescence
magnetic nanoparticles. The Morris Water Maze Task was performed 3 months after the
intracerebral injection or the last intravenous injection in order to measure spatial
learning and memory (Kim, et al., 2012).
For the intra-cerebral injections fur on the dorsal and ventral surfaces of the mice was
removed and hASCs were transplanted into both hemispheres of anaesthetized 11-month
old mice, each individual injection came from a single donor and injections over a
different period of time were from different donors (Kim, et al., 2012). In Vivo
florescence measurements were obtained 1, 3, 10 and 30 days post-injection and all
