Document Summary

Imin 405: techniques for the study of innate immunity. The path to discovery of new immune subsets. Appear to play a role in response to helminth infections. Potentially important in asthma, allergic airways disease, and the pathology of colitis. Type-2 immunity to helminths involves basophils, eosinophils, and mast cells as sources of type-2 cytokines. Look at cells producing il-13 (critical in response to helminth infections) Pcr - total cell population, no individual cells. In situ hybridization - individual cells; challenging technique. Elisa - total cell population; soluble proteins only. Elispot - spots produced by individual cells, but no identity. Western blot - cell lysate, no individual cells. Flow cytometry - individual cells in a mixed population; intracellular cytokine staining low sensitivity. Fluorescence microscopy - individual cells in a mixed population; intracellular cytokine staining low sensitivity. Transgenic mice - need to engineer/expensive to buy (done in this study)
