MCIM 224 Lecture Notes - Lecture 5: Bacitracin, Dna Supercoil, Semisynthesis
Document Summary
Goal - to provide all the factors (nutrients, temp, atmosphere, etc. ) necessary to obtain optimal bacterial growth. Incubators set at optimal growth temp (usually 37 c) Anaerobic jar chemical reaction removes o2 and generates h2 + co2. 1) some bacterial species cannot be grown on lab culture media. If they require unusual / unknown growth factors. If they occupy unusual environmental niches that can"t be duplicated in the laboratory. Only a small % of all microorganisms have been grown in the lab. 2) media can be formulated to favor growth of one species over another and/or visually distinguish one species from another ( selective vs. differential media) Has additives that are toxic to some organisms but not others eg. sabouraud agar - formulated with slightly acidic ph. Inhibits growth of bacteria, but not fungi. No inhibitors added (all species can grow) Differentiates between species based on appearance on media eg. blood agar (contains sheep red blood cells)
