In this laboratory you will isolate the salivary glands from larvae of the fruit fly drosophila virilis to observe their polytene chromosomes. It is possible to directly observe polytene chromosomes under a conventional light microscopy as they contain multiple strands of dna that are partially uncoiled. You will also assess the impact of temperature on the initiation of confined swelling or puffing. You will take microscope photos of your polytene chromosomes and measure their length and use this information to estimate the relative packing of dna within polytene chromosomes compared to conventional chromosomes. Theory: description and peculiarities of polyteny, morphology of polytene chromosomes, multistrandedness of polytene chromosomes, banding pattern within polytene chromosomes, puffing of polytene chromosomes, gene mapping. Pre-lab activities: carefully review these two videos (video 1 and video 2) illustrating the proper technique to dissect and clean up salivary glands from 3rd instar larvae of drosophila virilis.
