Review from last lec: proteins are part of signaling pathway. Purpose of signaling pathway: how cells communicating *with itself or with another cell. What is rational design vs. high throughput screening. At the end of the day, you want your drug to hit 1 specific target. You can choose the size, composition, acid/base properties, etc. This is important because you don"t want a deadly chain reaction to happen. It"s important to know as much as possible about what your target interacts with. 3 main inspirations for rational design: natural products: chemicals produced from nature (from bacteria, animals, plants, etc. , endogenous: (ex; atp, other protein binding partners) we can hijack the structure change it, design de novo: design molecule from scratch. Hardest way to make drugs, but sometimes you don"t have a choice. Tools typically used in rational drug design: x-ray crystallography: you grow a crystal of your molecule. Each part of this crystal has one protein unit.
