Ubiquitous throughout the body and not so similar to other proteins. Maps based on phenotypes do not cover enough of genome. Allows genes and mutations of interest to be identified more rapidly and easily. Determine physical location of large pieces of dna (large-insert clones) Used by collins for the human genome project. Make and sequence these segments then find the overlaps. This allows to create the entire segment - results in the contig. Take dna and break it up randomly into small fragments.
