PSL300H1 Lecture Notes - Lecture 23: Lipophilicity
Document Summary
Estrogen can bind to dna: high affinity. Two hormones can bind onto the same receptor, but one may bind with higher affinity: saturable. When all of the receptors are taken up, nothing else can bind: specific. More specificity means that it is harder to knock off the estrogen from the receptor: similar to affinity. Proper conformation is required for the ligand to fit into the binding site of the receptor: reversible. If a ligand comes off the receptor, another can come in and bind to that receptor. Hormone receptors and mode of action v peptide hormones: cannot penetrate target cell, bind to surface receptors and activate intracellular processes through second messengers v steroid hormones. Include cytosolic and nuclear receptors because lipophilic hormones can diffuse across the plasma membrane into the cell. Tend to directly alter gene transcription and have genomic effects: plasma membrane receptors.
