Hot to take pieces of dna and turn it into a bigger genome. Its is easy to generate data but putting it together is hard. Before it was hard to just generate the pieces. But now, it is hard to put the pieces together. Two pieces of dna came off of a sequencing read and they are 150nt. There is an overlap between the 2 pieces that is 25nt in length. The length of this contig is 275 nt in length. Can be confident to say that these two genes belong together: Chance of 1 side being identical is 0. 251=0. 25. Chance of 2 sites being identical is 0. 252=0. 0625. Chance of 25 sites being identical = 0. 2525 = 10-15. Don"t worry about what the actual repeats are. Because these are repeats the contig can be shifted. We don"t know how to position the contig because of this.
