Genetic mapping using marker alleles such as micro satellites. Forward genetics: trying to find what gene causes a certain phenotype (phenotype gene). Methods for this include positional cloning where you look for the specific gene responsible for a specific phenotype. Reverse genetics: trying to determine what the function of a particular gene is (gene phenotype). First, we try to determine whether the trait is dominant or recessive. We can determine this with a test cross, where the mutant phenotype is crossed with a wildtype phenotype. If the progeny is 100% wt, then the wt is dominant and the mutant is recessive (the case of the groodies). The progeny can also be 100% mutant (if dominant). Then we want to do a population screen to find the independently-derived mutants. These independent monogamous mutants can then be crossed together in a complementation assay.
