Suspected pathogen must be grown in pure culture: cells from pure culture of suspected pathogen must cause disease in a healthy animal. Suspected pathogen must then be re-isolated and shown to be same as original. Classified by sequencing rrna: bacteria, cell walls that contain peptidoglycan, archaea, cell walls when present are lacking in peptidoglycan, eukarya, includes protists, fungi, plants & animals.
