Document Summary

Transport vesicles: from a donor to a target compartment. Vesicle coatings: clathrin-coated vesicles: from plasma membrane and b/w endosomal and golgi compartments, copi(coat protein i)-coated vesicles: from golgi back to er, copii-coated vesicles: from er to golgi. Made of 3 polypeptide heavy chains and 3 polypeptide light chains called a triskelion. Requires adaptor proteins (different membrane = different adaptor protein) that cover the inner layer of the vesicle; (1) allows the binding of the clathrin coat to the membrane and (2) traps cargo receptors (adaptor proteins are cargo receptors specific) Binds to pip2 (phosphatidylinositol 4,5-bisphosphate) to activate the 2/ 2 binding site which attaches to cargo proteins inducing membrane curvature which then induces cooperative binding amplifying vesicle formation and vesicle budding. Shapes and pinches off vesicles via electrostatic interactions with the head groups. Protein that spirals around and pinches off the neck of budding vesicles being formed.
