Proteins can be modified by other proteins for a short term (kinases) or they can be long term such as nuclear pore formation. Genetic method based on modular structure of transcription. Binding of gal4 to promote induces transcription of a set of genes. Gal4 protein can be cleaved into 2 domains. Makes use of the fact that separate gal4 domains cant activate transcription. Protein of interest brings gal3 domains into close proximity for activation. Vector consturcts are intoroduced into cells that also have a reporter lacz gene. Mate both vectors (cloned gal3 domain with gene or protein of interest) together for screening. If proteins of interest interact with each other, transcription will occur and the reporter gene is expressed. If create library of yeast strain with different proteins and make a system, youll have certain proteins on your control and on your activity. Look for the colonies for which turns blue and which don"t.
