The microbial population will determine the shelf life of a food. It can be really hard to detect and enumerate bacteria strains. Culture methods: the cell has to form colonies on the plate or turn turbid in broth. Cellular component (artifact) methods: these detect components of the cell membrane such as proteins, fatty acids on the membrane, and antigens. Nucleic acid-based methods: these target dna and rna. Microcalorimetry- helps measure the amount of heat given off by microorganisms. Atp bioluminescence: how much atp is produced by a microorganism. Plate counts- colony forming units (cfu: growth of m/o on solid media, types: Pour plating (pour sample into dish, pour agar over sample and swirl: good method for microaerophiles because the bacteria tend to stay near the bottom fo the agar and can grow within low oxygen environments. Membrane filtration: this is good for samples that have low counts and are diluted.
