Heat-sensitive microbes are damaged and the actual appearance of colonies is not able to be done. At least 100ml of a relatively pure sample of water are filtered through a membrane and this filter is transferred to a petri dish where the bacteria is grown. Uses the fact that the greater the number of bacteria the more dilution is necessary to stop growth of bacteria and test tubes are counted to see how many bacteria are left. Measured volume of a bacterial supsnesion is placed within a defined area on a microscope slide. Number of bacteria can be counted in area are counted. Average of each viewing field is calculated and the average number per square centimeter is calculated. How is turbidity used to measure bacterial number. Spectrophotometer uses light and the amount of light that passes through the sample determines the amount in the sample.
