Study of microbial structure: the light microscopes, how to stain and prepare microbes for viewing, electron microscopy, tem-sem. Viruses and acellular agents: structure, types of viruses, how viruses multiply, use in biotechnology, viruses can infect bacteria and animals. Isolation of a pure culture: uptake of nutrients, media, culture methods, colony of bacteria arises from a single cell, different types of media are used to grow different bacteria. Microbial metabolism: atp, catabolism vs anabolism- things are always happening at the same time- things are being made and broken down. We want to understand big picture: biochemical pathways/cycles. Anaerobic respiration: microbes have about 20 metabolic pathways. Microbial genetics: gene structure, dna replication, transcription, translation, the genetic code, mutations, dna repair, conjugation, transformation, transduction, bacteria replicate by binary fission- need something to create genetic variability (conjugation, transformation, transduction- uses bacteria phages)
