Arise from certain scs at certain times, differentiate from each other. More potent- more different types of tissues they can become. Totipotent: responsible to extra embryonic tissues (placenta: only around for first few cells. Pluripotent: can become everything but extra embryonic tissues: lose ability, decrease potency. Oligopotent- can form a few tissue types within layer. Asymmetric cell division so never lose scs. Fertilized egg blastocyst (source of scs) form all of development until adult mouse. Icm in middle are pluripotent and form rest of mouse. Put on fibroblast feeder layer- other cells form mouse that form monolayer. Need stem cells to stay stem cells. If start to differentiate- then can"t be injected. Not possible to make synthetic media without feeder layer, but feeder layer is still standard process.
