Qualities of a research method technique: structure / function. Why is it important: static / dynamic. Invasiveness: risk and ethical concerns, scale (spatial resolution, speed (temporal resolution, cost and accessibility. Highly invasive methods: surgery with stereotaxic device. Infusion = puts chemicals (like drugs) directly in to local brain region. A microscopic approach: histology = microanatomy of cells. Tissue must be processed and stained to be visible: many types of microscopes. Golgi silver stain = random singe cells, structural features. Initially names the black reaction (la reazione nera) Cells are filled by microcrystallization of silver chromate. Darkly stains full neuron, useful for detailed single cell imaging. Stains fatty myelin sheath that forms insulation around axons. Dark coloring = axons, fiber tracts (white matter) Stains cell bodies of neurons by darkening rough endoplasmic reticulum material in nucleus. Useful for identifying populations of cell bodies and layered structures (e. g. cortex)
