A dna library is a collection of cloned fragments of dna, from a single source. Libraries derived from the genomic dna of an organism are called genomic libraries. Libraries derived from the coding regions of the expressed genes of an organism are called cdna libraries. Those are made of complementary dna (cdna) synthesized from rna. Genomic dna from a single individual is isolated and fragmented into smaller pieces. Recombinant vectors are transformed into bacteria that grow into colonies that collectively contain clones representing the entire genome. In theory, the library should contain all sequences from the genome in equal frequency. Reverse transcriptase is used to copy the mrna into a single-stranded. A primer complementary to the poly-a tail (oligo-dt) is used. After the first strand of dna is synthe-sized, mrna is enzymatically removed. The second strand of the dna is synthesized by dna polymerase, using the first strand as a template.
