The endocannabinoids are unusual because the act backwards, they are produced by the postsynaptic cells and effect the presynaptic cells. This action seems to be apart of a negative feedback cycle. This allows the postsynaptic cell to tell the presynaptic cell that too much of the neurotransmitter is being release enabling the process to be slowed. The synaptic vesicles do not contain the endocannabinoids but instead can contain any other neurotransmitter. As the cells become more depolarized voltage gated calcium channels on the postsynaptic cell will open. The calcium coming into the postsynaptic cell will stimulates the enzymes that produces the endocannabinoids. The endocannabinoids can then directly diffuse across the membrane because they are unsaturated long carbohydrates chains that are fat soluble.
