Obtain e. coli bacteria in a cell paste from a frozen preparation of cells: stir until a smooth suspension is achieved. Pipet 4. 5 ml of bacterial suspension in saline-edta in a blue top tube conical tube: this should be about 0. 35 g of e. coli. Pipet 0. 25 ml of lysozyme solution into the sample: mix by gently inverting the tube. Incubate this prep sample in a water bath at 37oc for 20 minutes: invert the tube every 5 minutes. Add 0. 5 ml of 10& sds: mix by gently inverting the tube for several minutes, suspension should become clear. Incubate prep sample for 10 minutes in water bath at 60oc. Let prep sample warm up to room temperature: about 10 minutes.
