Tolerance and dependence essentially develop together, treatment protocols (e. g. , opiates, no curative for substance abuse, chronic problem that you treat but can never cure, basic goals, reduce withdrawal symptoms. So getting off drugs easier: reduce craving, craving is a symptom that extends for a long time, block the effects of drugs. It will keep the drug from getting you high so there is no positive reinforcement. Lately we start to go into behavioral health, but it"s not very developed. You don"t get much of a high or none at all. Inconvenient: easily abused by increasing the dosage to get high, buprenophine, mu agonist / kappa antagonist, 1/2 life ~37 hours. Found post synaptic: d2 family, d2, d3, d4, gi (inhibitory) --> down regulation (decrease) in camp. Increase in k + conductance, opening potassium channels: pre and post synaptic, pre(autoreceptors) (not just on the axon terminal, but some can be found on the soma too)
