Document Summary

Cell signaling sense, receive, and respond to information from the environment. Paracrine ligands released from cell to receptors on nearby cell. Synaptic neuronal cells send signals down path. Variationpresence of two different receptors in two different cell types. Degree of receptor activation can produce differential response. Presence of same receptor, but different intracellular signaling components. Adrenalin binding to g alpha leads to plc activation pip2, ip3, Dag activation ca2+ activation smooth muscle contraction. Adrenalin binding to g beta leads to ac activation atp, camp activation heart muscle contraction. Transfect target cell lacking receptor for ligand w/ cdna expression vector pool; one cdna encodes receptor; cells transfected will express receptor; identify cells by labeling ligand w/ fluorescent label; once identified, cdna for receptor cloned out and sequence determined. Kd = ligand concentration at which half of receptors are bound to ligand. Lower kd = higher molecular complementarity = higher affinity.
