Inheritance of alzheimer"s due to mutations in genes: app gene - early-onset - abnormal processing of app may directly cause. Alzheimer"s in certain families: ps1 & ps2 genes - early-onset - another gene that may be directly causative of alzheimer"s, apoe gene - late-onset - gene that may contribute to getting alzheimer"s at older age. Individuals or cells expressing the "swedish mutation" (found in large swedish family) in app over-produce abeta: presence of swedish mutation increases deposits of ab, neurofibrillary tangles! What causes those: mostly we do not know, apoe, however, we do know that certain forms of a cholesterol carrier protein called apoe can strongly predispose individuals to getting alzheimer"s.
