A synaptic vesicle on its own is a very complex structure. New vesicles made in the cell body and then are trafficked via kinesin and microtubule system to the nerve terminal. If you mess up the kinesin then u mess up the vesicle transportation (the proof) They don t stand still and they move in straight lines. We bleach (the black bar) but now with myosin ii manipulated the black bar doesn t fill as fast and doesn"t completely fill thus we can deduce that vesicle mobility depends at least partly on myosin ii. Summary of today"s class: movements depend on actin and myosin. Its not just there to hold the structure but are dynamic and allow vesicle to move. And not to just move in straight line. Red is the control, pink is a knock down of myosin. Thus you can see when you have a mutation of myosin ii then there is less of a recovery.
