Bright field microscopy: stain it and kill it. Phase contrast: puts halos around the specimens, dark is darker and light is lighter. Subcellular fractionation: from cells to organelles: step 1: homogenization, 4 ways: sonication needle, force it through small hole, Detergent and shear cells with rotating plunger: step 2: isolation, centrifuge till you get the one you want, step 3: purify, through velocity centrifugation and then equilibrium centrifugation. Spicing: 1 gene can make 1 pre-mrna, then it"s processed to make several mature mrna and that makes several proteins.
