Cytology
Cells/nuclei come in all shapes and sizes
- top left corner
o nucleus is in center
- bottom left
o pink is cytoplasm
o nuclei are basal (located at bottom of cell)
- top right
o skeletal muscle
o dark dots are elongated nuclei
o basophilic staining nuclei
o nuclei located towards periphery of cell
- bottom right
o pink dots = RBCs
o kidney bean shaped nucleus
Parts of a Cell

o stained with crystal violet (basic stain)
o nucleus looks like owls eye
o you can see the nucleolus in the middle of nucleus
o at periphery of nucleolus we have a pale area, which is the nucleus
o this is where the inner and outer nuclear membrane would
be (cant see it in light microscope – only in EM)
o intermembrane space of nuclear membrane is continuous
with endoplasmic reticulum
o the green outline is the cell membrane
o note: we can only see the plasma membrane on electron
microscopy

o nerve tissue stained with H&E
o the bracket is the cytoplasm
o the nucleus is surrounded by the nuclear membrane with nucleolus floating in middle
Cellular Features
- euchromatin
o loosely coiled
- heterochromatin
o called peripheral heterochromatin
o peripheral heterochromatin is on the side of the nucleus so it also
helps highlight the nuclear envelope
o tightly coiled
o darker stained when stained with a basic dye (ex: hematoxylin)
- image is the liver
- if we see this patchy appearance in the cytoplasm, it’s the glycogen (bc euchromatin and
heterochromatin are specific to the nucleus)
- nissl bodies
o stained with tolydyne blue (basic stain)
o represent rough ER and the free ribosomes within the cytoplasm
