Mitochondria: main role to make atp and determine fate of cell. Culturing preserves function and survival of a cell. Also allows us to isolate specific organelles or proteins. Culture technique grows cells or tissues outside the organisms under controlled conditions. Cells are isolated by breaking down the cell-cell and cell-matrix interactions. Then cells are supplied with nutrients (aas, vitamins, growth factors, serum - blood derived product) and kept at 37 in co2 incubator. Growth factors act as mitogens give signals to cell to divide into colonies. Co2 reacts with salt in the media to buffer the ph. Cells can be cultured through suspension (unicells) or adherent solid surfaces. Tripson: enzyme that cleaves all the proteins on the outside of the cell. Edta: sucks up ions (ca and mg) that bind to certain proteins outside cells that make sticky proteins more sticky. Scoops up ions to make protein surface less sticky.
