Gene therapy uses delivery of dna into cells. Dna libraries (the bigger the insert, the fewer clones you need: cosmids, phage, phage display, bacs (bacterial artificial chromosome, yacs (yeast artificial chromosome) Phage : bacterial virus that infect e. coli (and multiplies in it until it dies) There is a restriction site at each end of the stuffer sequence. Digested stuffer dna can be removed physically by a variety of methods (use pcr to insert restriction sites into dna to cut the stuffer sequence and then ligate). When you blot phage, you do it on a lawn of bacteria, and the next morning where there are holes on the agar plate where the cells have disintegrated due to the phage. Called a plaque: lawn : when all the individual colonies on a petri-dish agar plate merge to form a field or mat of bacteria. Cosmids (cos sites + plasmids) new, bigger, improved (plasmid library with a phage step before)
