BIOLOGY 242 Midterm: Lab 13
Document Summary
Effectively stains endospores of bacteria and ascospores of fungi. E(cid:374)dospores are highly resista(cid:374)t a(cid:374)d are produ(cid:272)ed (cid:271)y (cid:862)lo(cid:449) g+c gra(cid:373) positi(cid:448)e(cid:863) (cid:271)a(cid:272)teria including bacillus sp. and clostridium sp. Once endospores are stained they resist decolorization: they are stained by being heated/steamed in the presence of malachite green dye, vegetative cells are counterstained with safranin. Endospores can be: shape: spherical or oval, location: central, terminal, or subterminal, size: bulging or non bulging. Add more dye as needed, making sure that the paper does not dry out: remove the filter paper and decolorize by rinsing with water, counterstain with safranin for 1 minute, rinse with water. They have multiple layers: outer protein coat (chemical/enzymatic resistance), cortex-thick layer of peptidoglycan (temperature: list 3 human diseases caused by bacteria in the genus clostridium. Include the disease and the name of the organism(s) that causes it.
