E1 is the enzyme that performs the oxidative decarboxylation of pyruvate, e2 performs the transfer to coa and e3 regenerates the cofactors in order to reset the cycle. Stoichiometric cofactors which consist of : coa and nad+, these enzymes are equivalent to a substrate and are consumed during the reaction. Enzyme e1 is a key site of regulation.
