Plasmids are not essential to bacteria but if bacteria picks it up it is helpful. Cell wall is important because it provides strength, rigidity and protection. Cell wall of bacteria is composted of peptidoglycan unlike an archaea. It"s structure; cytoplasmic membrane and on top they have a thick, dense layer of peptidoglycan on top. Thin layer of peptidoglycan compared to gram positive bacteria. You use a series of dyes to stain cell to make them visible (crystal violet) Add second stain (iodine) helps to fix crystal violet in cell configuration. If positive it wont be readily washed away because alcohol cannot get into pepti. To clean the cells because the layer is so thick. Weakening the cell wall can lead to cell rupture and death. A layer that surrounds bacteria and rests on top of cell wall. Made up of polysacs, helps protect from extreme heat, desiccation, external environment and invaders.
