BIOL 2021 Chapter Notes - Chapter 2.1: Fas Ligand, Death Domain, Fas Receptor
Document Summary
There are both extrinsic (from outside) and intrinsic (from inside) pathways. Death receptors/tnf receptors trigger the extrinsic pathway of apoptosis: transmembrane proteins containing an extracellular ligand-binding domain and an intracellular death domain. Tumour necrosis factor (tnf) is a family of signal proteins: ex. Fas ligand: membrane bound protein on a target cell: domain. When activated by the binding of a fas ligand, the death domains on the fas receptor bind to intracellular adaptor protiens: fadd is the adaptor protein and binds to initiator caspases when activated. Disc (death inducing signaling complex): fadd and caspase complex collectively. Executioner caspases can then cleave targets to begin apoptosis. Also called the mitochondrial pathway of apoptosis. Can occur due to injury, stress, dna damage, or the loss of survival signals. Initiator caspases bind to the apoptosome, cleave and activate each other, and a downstream cascade occurs, leading to apoptosis. Bcl2 are a family of proteins that control the.
