Pip3 recruits 2 protein kinases to membrane via ph domains- akt (protein kinase: & phosphoinositide-dependent protein kinase 1 (pdk1). Figure 15-53 one way in which signalling through pi 3-kinase promotes cell survival. Extracellular signal activates rtk, which recruits & activates pi 3-kinase. Pi 3-kinasse produced pi(3,4,5)p3, which serves as docking site for two serine/threonine kinases with ph domains- akt & pdk1- & brings them into close proximity in plasma membrane. Akt is phosphorylated on serine (by third kinase) which changes conformation & allows akt phosphorylation on threonine by pdk1 which activates akt. Activated akt dissociates from plasma membrane & phosphorylates target proteins. Ex: bad protein hold inactive apoptosis inhibitory protein.
