Good sterile technique is always the best defense against contamination. Fungal/yeast inhibitors (only used on longterm cultures) Because bacteria grow rapidly, antibiotics are used for all sample types. Since fungi & yeast grow more slowly, antifungal agents are usually only used on long-term cultures (amnios and solid tissues) because they have a detrimental effect on growing cells. Suspect if chromosome breakage exceeds background rate for lab. Unstable amino acid that breaks down to d-glutamine upon storage, which cannot be used by cells. Must be added to media immediately prior to use: mitotic stimulants/mitogens. Needed for some, but not all cell types to encourage cell division. Causes a wave of cell division at 48 hours, and then at 24 hour intervals. Darker staining, more condensed metaphases are older and more mature (can sometimes be interphases)
