Lecture 13: biotechnology
Transcriptomes
- The set of RNA molecules produced by an organism, organ or group of cells
- Defines the genes that are being expressed
- Transcriptomics= study of
transcriptome (complete set of RNA
transcripts that are produced by the
genome under specific circumstances
or in a specific cell
Blue= under expressed relative to others
Red= over expressed
- Can see what genes are changing
Restriction enzymes= endonuclease
- Bacteria are attacked by viruses called bacteriophages
- Bacteria defend themselves against viral DNA by cutting foreign DNA with restriction
enzymes
- Eah estitio eze uts at a speifi seuee aset i the hosts genome

Some common restriction enzymes
- They are read as palindromes
(read same both ways)
Gel Electrophoresis
- Gel electrophoresis tank (in
here we have some agar (like a
jelly). At one end there will be
wells and in each of those, we
stick DNA)
- Apply electric current
- DNA will be attracted to positive end (they
are negatively charged)
- On one side, the same piece of DNA has
been cut with restriction enzymes
- Single piece of DNA that has been extracted out of an animal

- if there is a mutation, then the enzyme will not cut at that site
D + E = 973 bp (longer)- instead of having 2
bands (D and E), there would only be one band
further up at 937bp
- We can cut DNA but also join them back
together using DNA ligase
