Document Summary

At morula stage blastomeres make their first decision. Inner cells are more likely to become icm and outside cells to become trophoblast. Aside from promoting trophoblast genes, cdx2 also repressing other genes of cell type, like icm genes. Cdx2 mutant embryos die around the time of implantation. Those cells die without the expression of cdx2, therefore cdx2 is essential for segregation of icm and te lineages at the blastocyst stage by ensuring repression of oct4 and nanog in the te. Oct 4 is required for icm, if removed it you get te. When embryo is cdx2 recessive, the polarity is messed up in the late stage of blastocyst. Rtcpr: can tell you levels of rna and wild type and mutant background. Use of immunos to show the missing expression of cdx2 in cdx2 mutant embryo. Oct4 and nanog are expressed in outer cells of cdx2 mutant blastocyst.
