A protease will cleave a section of the n-terminal portion (in the extracellular environment), allowing the ligand to bind and activate the receptor, causing a intracellular signal. Ligands: some ligands need a trigger to become active. They might be close to the receptor already, awaiting to be enlightened! For example, the rhodopsin receptor, once bound to retinol, will change its conformation: other ligands come from other cells, they need to travel a certain distance, coming from blood stream for example, before reaching target tissues or cells. Class c gpcrs: sushi domain creates disulfide bonds required to maintain tertiary structure: keeps alive a binding site! Ligands can bind to multiple different sites on a receptor depending on the class or the type of receptor.
