Document Summary

Lymphocyte development, negative regulation & immune tolerance (module 4) Lecture 35 immunotherapy, immuno-oncology with special guest dr. caroline lambert. Lecture 36 autoimmunity, multiple sclerosis with special guest sarit klein. Lecture 37 allergies and mini wrap up. Positive selection determines mhc restriction: done by cortical epithelial cells in the thymus which are insensitive to radiotherapy. Negative selection ensures self-tolerance: mediated by bone marrow derived cells in the medulla of the thymus which are sensitive to radiotherapy. Thymocytes whose tcr shows too much reactivity for self-peptide:mhc complexes are deleted (die of apoptosis) Aire is a protein (transcription factor) that induces the expression of many tissue-specific proteins in the thymus: source of peptides on which to test tolerance to self-peptide:mhc complexes. Prof wld like us to try to explain the experiment (top figure) on our own time. Cortical epithelial cells positive selection, not radio-sensitive. Medullary epithelial cells negative selection, originate from bm precursors, radio- sensitive.
