Immune system of mouse-----has been characterized more extensively than any other species. Mouse-model has an enormous impact on clinical intervention in human disease. Inbred mice: by inbreeding for 20 generations ---inbred strain. Hematopoietic cells can be removed by high dose of x rays. Allow replacement of donor cell from another animal, to examine development of lymphocyte and their functions. Hematopoietic stem cell can be obtained from bone marrow, umbilical cord bld or peripheral bld after donor is trted with hematopoietic growth factor (ex. To investigate importance of tc / effect of lack of tc / study tc subpopulation in mice with no tc of their own. Method: surgical removal of thymus at birth (since mature tc only occurs after birth) This won"t deplete all t cell ---- still can find some in bone marrow and lymphocyte (tc is not expressed only from bone marrow) Purpose of depletion: in order to put the cell i want to investigate.
