Transport to the er is co-translational (i. e. transport is occurring while translation is in progress) Pathway also known as secretory pathway because secreted proteins are synthesized in er before vesicle-based transport carries them out of cell. Bound by single membrane (which is continuous with outer membrane of nucleus) Punctate black dots on rough er represent membrane-associated ribosomes that are synthesizing secretory proteins (red) These proteins travel through golgi apparatus & in vesicles to reach outside of cell. Note: associated ribosomes on cytosolic side of er membrane. Recall: known as rough er because of punctate appearance created by associated ribosomes. Both associated & free ribosomes are translationally active & come from same common pool; they cycle on & off er membrane. Question: are proteins transported to lumen of er immediately upon translation. Hypothesis: if transport occurs during translation, then proteins will exit the ribosome & go directly into a microsome.
