Rna pol recognizes a certain sequence located 35 bp upstream of +1 - rna recognition site, It binds to another specific sequence rna binding site located at the -10 element position 5"- Tataat-3" also called pribnow box or -10 element. Both sequences are on coding strand - complementary to template strand. They are consensus sequences - most common promoter sequence. Even if -10 element is intact - if -35 element isn"t - transcription wont happen. The sigma factor positions the holoenzyme at the -10 element and core enzyme starts to unwind.
