True motility -> moves across the microscopic field (usually zig zagging its way along) Brownian motion -> stay in place but vibrate or shake (due to water molecules bombarding organism and is not characteristic of true independent movement) Spread petroleum jelly around concavity of depression slide (4 dots) Transfer loopful of water to centre of coverslip, then transfer loopful of culture in drop and let it enter water without agitating (will ensure flagella remain in tact) Lower depression slide with concavity facing down onto coverslip. Turn depression or slide so the specimen will hang from coverslip to depression.
