Cancer progression occurs because of defects in molecular machinery and regulatory mechanisms that normally control cell shapes, functions, interactions and numbers. = own machinery going awry (different from external bacterial infection) Different pathways affected that can lead to cancer. Can have problems in cell adhesion (cadherins) Can have problems in pathways that control cell numbers (proliferation/death) There are ~30-100 trillion cells in a human body. Billions of cells likely experience mutations every day. Most mutations are fixed (dna repair machinery) If mutations not fixed proteins like p53 kill off dangerous cells. = cells w/ persistent mutations are normally destroyed. Cancer develops from one of these cells gaining a mutation that allows it to survive and divide forming a tumour. Cancer develops from one cell that divides to form a tumour. As single original cell keeps dividing, unconstrained growth, forms deadly tumour in less than 40 cell doublings.
