Difference in gene expression between two cells is small; differences in gene expression may reflect differences in underlying cellular state. Snapshot of transcriptome in particular moment in time. Sample dna bind to microarray based on complementarity. Relatively unstable, so converted to cdna (using reverse transcriptase) Label entire cdna in one population from one sample with one colour, and other population in another colour. Completely hybridize (bind) both populations of cdna against the array. Unbiased wrt hypothesis, e. g. don"t assume gene x is involved in disease. What pattern of gene expression best defines patient-ness . Is gene expression pattern more like health or disease. Inactivation: active drug is converted into inactive products. Maintenance of activation: active drug converted into different but still active metabolite. Effect of cyp2d6 enzyme on analgesic effect of codeine. Cyp2d6 poor metabolizers will show poor analgesia from codeine. Microarray contains common variants of known pharmacogenetically relevant genes.
