Biology 2382B Lecture Notes - Lecture 2: Phase-Contrast Microscopy, Bright-Field Microscopy, Dichroic Filter
Document Summary
Imaging is very important for research in cell biology; it allows visualization of cellular and molecular processes. It was the creation of a light microscope that gave rise to the field of cell biology, and for a long while, light microscopy was a key tool of cell biologists. Coining of the term cell was done by robert hooke in 1665 when looking under a microscope. With improvements in microscopy techniques, cell biology is revolutionized. Anton was great with glass and was able to build many simple microscopes or microscopes with a single lens. With his fine handiwork, he was able to craft glass lenses which could achieve a magnification of 200x. As a result, he was able to observe living protozoa and bacteria, which he called animalcules . Features of a modern compound microscope (bright-field microscopy) Compound microscopes include bright-field microscopy which uses white light. The magnification of a typical light microscope is 40-1000x.
