Document Summary

One way to measure respiration is by looking at the chance oxygen concentration. You can use an oxygen electrode, which is a sealed chamber lled with puri ed mitochondria liquid. You can look at the change in [o2] because its not coming from outside. You want to know how the concentration changes. Grind up chlamy cells, brake the cells up, but you want intact mitochondria. Purify intact mitochondria and incubate them in a buffer so they don"t get destroyed, and put in electrode chamber. The [oxygen] in the chamber is going down: its oxidative phosphorylation because you"re oxidizing nadh. You don"t necessarily need o2 involved: if you don;t have adp and pi in the system, then you"re just oxidizing nadh. and conservation of energy in the electrons is through proton pumping. If you add adp and pi, you"re releasing the gradient and the rate of et goes up.
