Lecture 6: dna replication and recombination: antibody: high af nity, high speci city. The antibody-antigen af nity has to be high to keep the antigen rmly bound to hold pathogens. High speci city because the surfaces t perfectly: this high af nity and high speci city can be used to pull out one particular protein from a complex mixture. Lowering the ph would denature the antibody, so the shape is changed. The protein recognized by the antibody is then released and eluted: gel electrophoresis: employs the differences in mass and charge to separate. Dna has uniform charge per unit mass, as all charge comes from phosphate groups. However, with other proteins, they have different charges, and different shapes that affect how it diffuses through the gel matrix. There is a way to make the proteins diffuse solely by mass: sds- Proteins are soaked in detergent, and the protein is denatured and becomes noodle-like.
