Lab 1 – September 15, 2011
Sterile technique – keep plates upside down to prevent anything falling in
Link between genotype and phenotype
Prototroph: cells able to make own amino acids/macromolecules
Autotroph: cells that require growth medium to be supplemented with compounds they
are unable to synthesize
Genetic nomenclature
-leu
oformat is lower-case, italicized
oleucine gene mutated => leucine autotroph
cannot produce leucine
leucine must be in medium
-lac
obacteria cannot use lactose as carbon source
ocarbon in medium, no growth
Carbon sources for bacteria
- glucose
- lactose
- maltose
- galactose
- sorbitol
MacConkey plates: indicator plates to determine if bacteria can use carbon source
present
- colonies red => can use carbon source
- colonies white => cannot use carbon source
- colour changes based on pH
