Anesthetize the animal--put is in the stereotaxic apparatus--cut open the scalp--drill the holes--insert the electrode or cannula--and lower it to the proper depth. A subcortical lesion is made under the guidance of a stereotaxic apparatus. The coordinates are obtained from a stereotaxic atlas, and the tip of an electrode or cannula (small metal tube) is placed at the target. Radio frequency lesion method lesion is made by passing radio frequency current through the electrode (stainless steel wire that is coated with an insulating varnish). It destroys all brain tissue near tip of electrode by producing heat (rf) destroys: neural cell bodies and axons of neurons that pass through this region. Injection of molecules of toxic saporin attached to an antibody (conjugate attach together) targeted against a protein found in particular neurons in the brain produces even more specific lesions.
